Abstract

A program in BASIC is proposed for ‘off-line’ optimization of experimental parameters in HPLC by the Simplex sequential method. The program is modular and permits adaptation of the chromatographic resolution definition to the user’s needs with a minimum of programming knowledge. The program is applied to improve separation of 2,4-dinitrophenylhydrazones of isolated carbonyl compounds in beer. The optimized parameters are: flow, initial methanol concentration, concentration gradient, and initial isocratic time.
